I don't use ZBrush but I use mac, so if it's the Mac and not Zbrush try the following:

Run and repair your permissions of the HD. Go to Macintosh HD:Applications:Utilities:Drive Utility and select the drive, then select Repair Permissions.

Is your HD the only HD connected to the system? If no disconnect the other HD's and reboot with them disconnected and try to save.

Are you using an administrative account? If you are using a standard account you may need to create an administrative account,
If you are using an administrative account drag zbrush folder to desktop and run it from there, if that fails try and create another administrative account and see if that works.

Remove your zbrush preferences from Macintosh HD:Libraries:Preferences and Macintosh HD:User
    your user name:Preferences
and put them on your desktop. If you have them both in Libraries and User put them in your desktop in different folders so if you need to put them back you know where to find them. I don't know what the file names for the preferences for zbrush are called, they could be anything from "com.autodesk.zbrush" to "zbrush.pref" to anything else. but whatever yo do MAKE SURE YOU ONLY REMOVE THE ZBRUSH PREFERENCES, NOTHING ELSE.

Also where is your scratch disk mapped to?

    Macintosh HD:User:Username:Preferences

becomes username raspberry for some reason. Yeah forums again!
